The Spanish Inquisition had been established in 1478, particularly with the goal of investigation Jewish conversos -- those who had converted to Christianity -- to make sure they truly were living and believing as Christians, not reverting to Jewish practices.
By 1492, the anti-Jewish measures were stepped up further and all Jews were given the order that they had to be converts to Christianity or would be forced to leave Spain. This is known as the Alahambra Decree, by King Ferdinand and Queen Isabella.
During those years, hundreds of thousands of Jews experienced either forced conversion or explusion from Spanish territory.
